Setting fuel pressure question

Do I need to set the base line fuel pressure with the sled running ie: the 5lbs or is this a static pressure with the fuel lines plugged off and the sled not running??

You'll see the same pressure either way.

Really, I guess that makes sense. Will fuel pressure at idle affect air fuel ratios at WOT? Thanks
