Yes. Difficult but not impossible to do this way if you mean you are trying to insert needle between inner race of bearing and seal. Often, it's easy to damage seal. It can be done but seal lip is in your way.
Better to buy a good dental pick (mechanics pic) and pick the seal out by inserting pick in between the outer race of bearing and edge of seal. I have a pick that works great. At least that way, if you do happen to damage the seal edge a little, it's not as critical because the seal and OD of the bearing are stationery anyway so grease won't be flying out.

I have over 10,000 on the original bearing by using the pick and WD40 and a little compressed air with new grease yearly. This year I damaged my seal but discovered the Kimpex 6205 Bearing seal fit's perfectly on both bearings Drive and Jackshaft. Excellent information on this Forum has saved me hundreds of dollars.Yes. Yep, greased my bearings with a needle for years until I learned that most bearings have a removable seal.
Use to damage too many seals with that old method so I stopped using that needle method, plus if you pop that seal off you can get to 100% of that bearing for a much more thorough grease application, I think.
